Viruses in the genus Begomovirus have two genomic components of circular, single-stranded DNA, are transmitted by whiteflies and infect dicot plants. Although the economic importance of begomoviruses in soybean in Brazil is secondary, the detection of these pathogens in the crop is concerning. Samples of soybean and associated weeds showing begomovirus-like symptoms were collected in Paraná state. Infection was confirmed by PCR, and two putative nove I specles, provisionally named "virus A" and "virus B", were detected. The objective of this work was to characterize these two putative new species. Viral genomes were amplified using the DNA polymerase from phage phi29, cloned and sequenced. The complete DNA-B sequence of "virus A", and a partial DNA-A sequence of "virus B", were determined, Sequence comparisons and phylogenetic analyses indicated that the DNA-A from "virus B" and the DNA-B from "virus A" have the highest identity with Sida micrantha mosaic virus (SimMV) isolates A2 and B3, respectively. However, identity levels are below 89%, indicating that both viruses actually comprise novel begomovirus species. |
